HIF-1α Gene Interference Alleviates High Glucose Induced Apoptosis, Inflammation and Extracellular Matrix Deposition in Renal Tubular Epithelial Cells

摘要: 目的 探讨缺氧诱导因子 1α (hypoxia inducible factor 1α, HIF-1α) 基因干扰对高糖 (HG) 诱导 的肾小管上皮细胞 (HK-2) 凋亡、 炎症和细胞外基质 (ECM) 沉积的影响。 方法 将 HK-2 细胞分为 4 组: 对照组; HG 组; HG + shRNA-NC 组; HG + HIF-1α-shRNA 组。 按分组处理细胞后, 实时定量聚合酶链 反应检测 HIF-1α 和中性粒细胞明胶酶相关脂质运载蛋白 (NGAL) 的表达水平; 细胞计数试剂盒 8、 流式 细胞术和酶联免疫吸附试验分别检测细胞增殖、 凋亡和炎性因子的含量; 免疫印迹检测蛋白表达。 结果 与 HG 组相比较, HG + HIF-1α-shRNA 组 HIF-1α 和 NGAL mRNA 和蛋白表达水平降低, 细胞增殖倍数升高, 细胞凋亡率降低, 白细胞介素-6 (IL-6) 和肿瘤坏死因子-α 水平降低, IL-10 水平升高, 组织金属蛋白酶抑 制物 l 和 I 型胶原蛋白表达水平降低, 基质金属蛋白酶 9 蛋白表达水平升高 (P< 0. 01)。 结论 HIF-1α 基 因干扰缓解 HG 诱导的 HK-2 细胞凋亡、 炎症和 ECM 沉积。

Abstract: Objective To investigate the effect of hypoxia-inducible factor 1α (HIF-1α) gene interference on high glucose (HG) -induced apoptosis, inflammation and extracellular matrix deposition in renal tubular epithelial cells (HK-2). Methods HK-2 cells were divided into 4 groups: control group, HG group, HG + shRNA-NC group, HG + HIF-1α-shRNA group. The expression levels of HIF-1α and neutrophil gelatinase-associated lipocalin (NGAL) were detected by quantitative real-time polymerase chain reaction. The cell proliferation, apoptosis and the levels of inflammatory factors were assayed by cell counting kit-8, flow cytometry and enzyme-linked immunosorbent assay, respectively. The protein expression levels were detected by Western blotting. Results Compared with the HG group, the mRNA and protein expression levels of HIF-1α and NGAL were decreased in the HG + HIF-1α-shRNA group, the cell proliferation rate was increased, the apoptosis rate was decreased, the levels of interleukin-6 (IL-6) and tumor necrosis factor-α were decreased, the level of IL-10 was increased, the expression levels of tissue inhibitors of metalloproteinase l and collagen type I protein were decreased, and the expression level of matrix metalloproteinase 9 protein was increased in the HG + HIF-1α-shRNA group (P< 0. 01). Conclusion HIF-1α gene interference alleviates HG-induced apoptosis, inflammation and extracellular matrix deposition in HK-2 cells.
