敲低 CXCL8 抑制口腔鳞癌的细胞增殖和转移

Knockdown of CXCL8 Inhibits Cell Proliferation and Metastasis of Oral Squamous Cell Carcinoma

摘要: 目的 探究在 CXC 基序趋化因子配体 8 (CXC motif chemokine ligand 8, CXCL8) 在口腔鳞癌 (oral squamous cell carcinoma, OSCC) 中的作用。 方法 实时荧光定量 PCR (quantitative real-time PCR, RT-qPCR) 和 Western 印迹检测 OSCC 组织和细胞中 CXCL8 的表达; 分析 CXCL8 表达与 OSCC 患者临床病理特 征的相关性; 细胞计数试剂盒 8 (cell counting kit-8, CCK-8), 克隆形成、 流式细胞术、 划痕实验、 Transwell 实验和 Western 印迹检测 CXCL8 敲低对细胞增殖、 凋亡、 迁移、 侵袭和上皮间质转化 (epithelial-mesenchymal transition, EMT) 的影响。 结果 CXCL8 在 OSCC 组织和细胞中的表达显著上调。 CXCL8 高水平 与肿瘤大小、 TNM 分期和远处转移显著相关。 CXCL8 的敲低可以抑制 SCC9 细胞增殖、 迁移、 侵袭和 EMT, 促进细胞凋亡。 结论 CXCL8 在 OSCC 中高表达, 敲低其表达可抑制 SCC9 细胞的增殖和转移。

Abstract: Objective To investigate the role of CXC motif chemokine ligand 8 (CXCL8) in oral squamous cell carcinoma (OSCC). Methods The expression level of CXCL8 in OSCC tissues and cells was detected by RT-qPCR and Western blotting, the correlation between CXCL8 expression level and the clinicopathological features of OSCC patients was analyzed. The effect of CXCL8 knockdown on cell proliferation, apoptosis, migration, invasion and epithelial-mesenchymal transition (EMT) was detected by cell counting kit 8 (CCK-8) method, colony formation assay, flow cytometry, wound healing assay, transwell assay and Western blotting. Results CXCL8 was significantly up-regulated in OSCC tissues and cells. The high expression level of CXCL8 was significantly correlated with tumor size, TNM staging and distant metastasis. CXCL8 knockdown can inhibit the proliferation, migration, invasion and EMT of SCC9 cells, and promote the apoptosis. Conclusion CXCL8 is up-regulated in OSCC, and the suppression of its expression can inhibit the proliferation and metastasis of SCC9 cells.
