肉桂醛调节 IL-6 / JAK2 / STAT3 信号通路对变应性鼻炎大鼠鼻 黏膜细胞凋亡的影响

Effect of Cinnamaldehyde on Apoptosis of Nasal Mucosa Cells in Rats with Allergic Rhinitis by Regulating IL-6 / JAK2 / STAT3 Signaling Pathway

摘要: 目的 探讨肉桂醛对变应性鼻炎 (allergic rhinitis, AR) 大鼠鼻粘膜细胞凋亡的影响及机制。 方 法 构建大鼠 AR 模型, 并随机分为 AR 组、 肉桂醛低、 中、 高剂量组和氯雷他定组, 另选 10 只大鼠为假 手术组 (sham 组)。 检测大鼠血清 IL-6 水平、 鼻粘膜组织细胞凋亡和 IL-6、 JAK2、 STAT3 mRNA 表达以及 JAK2、 p-JAK2、 STAT3、 p-STAT3、 Bcl-2 及 Bax 蛋白表达。 结果 与 sham 组比, AR 组大鼠血清 IL-6 水平、 鼻粘膜组织 IL-6 mRNA 及 p-JAK2 / JAK2、 p-STAT3 / STAT3 和 Bcl-2 蛋白表达均升高 (P< 0. 05), 细胞凋亡 指数 (AI) 和 Bax 蛋白表达降低 (P< 0. 05); 与 AR 组比, 肉桂醛低、 中、 高剂量组和氯雷他定组大鼠血 清 IL-6 水平、 鼻粘膜组织 IL-6 mRNA 及 p-JAK2 / JAK2、 p-STAT3 / STAT3 和 Bcl-2 蛋白表达均降低 ( P < 0. 05), AI 和 Bax 蛋白表达升高 (P< 0. 05)。 结论 肉桂醛可促进 AR 大鼠鼻粘膜细胞凋亡, 其机制可能与 抑制 IL-6 / JAK2 / STAT3 通路有关。

Abstract: Objective To investigate the effect of cinnamaldehyde on apoptosis of nasal mucosa cells in rats with allergic rhinitis (AR) and its mechanism. Methods The AR model was established in rats and the rats were randomly divided into AR group, low-, medium-and high-dose cinnamaldehyde groups and loratadine group, another 10 rats were selected as the sham operation group (sham group). The apoptosis of nasal mucosa cells, the level of serum IL-6, and the expression levels of IL-6, JAK2, STAT3 mRNAs and JAK2, p-JAK2, STAT3, p-STAT3, Bcl-2, Bax proteins were detected. Results Compared with the sham group, the level of serum IL-6, the expression levels of IL-6 mRNA and P-JAK2 / JAK2, P-STAT3 / STAT3 and Bcl-2 proteins of nasal mucosa cells were increased in the AR group (P < 0. 05), while the apoptosis index (AI) and Bax protein expression level were decreased (P< 0. 05). Compared with the AR group, the level of serum IL-6, the expression levels of IL-6 mRNA and P-JAK2 / JAK2, P-STAT3 / STAT3 and Bcl-2 proteins in rats nasal mucosa cells in the low-dose, medium-dose and high-dose cinnamaldehyde groups and the loratadine group were decreased (P< 0. 05), while the AI and Bax expression level were increased (P< 0. 05). Conclusion Cinnamaldehyde can promote the apoptosis of nasal mucosal cells in AR rats, and the mechanism may be related to the inhibition of IL-6 / JAK2 / STAT3 signaling pathway.
