蜘蛛香总黄酮调控 miR-4429 影响胃癌 AGS 细胞的增殖、 迁移及侵袭

Effect of Total Flavonoids from Valeriana Jatamansi Jones on Proliferation, Migration and Invasion in Gastric Cancer AGS Cells via Regulation of miR-4429

摘要: 目的 探讨蜘蛛香总黄酮 (total flavonoids from Valeriana jatamansi Jones, TFV) 对胃癌 AGS 细胞 增殖、 迁移及侵袭的影响及其可能的作用机制。 方法 体外培养人胃癌细胞 AGS, 随机分为 Con 组、 TFV-L 组、 TFV-M 组、 TFV-H 组、 miR-NC 组、 miR-4429 组、 TFV + anti-miR-NC 组、 TFV + anti-miR-4429 组。 qRT-PCR, CCK-8 法、 平板克隆形成实验、 划痕实验和 Transwell 实验分别检测 miR-4429 表达, 细胞增殖、 克隆形成能力、 细胞迁移和侵袭情况; Western 免疫印迹检测 E-cadherin、 N-cadherin 蛋白表达量。 结果 与 Con 组比较, TFV-L 组、 TFV-M 组、 TFV-H 组细胞 E-cadherin 蛋白水平、 增殖抑制率和 miR-4429 的表达 量升高 (P< 0. 05), N-cadherin 蛋白水平和划痕愈合率降低 (P< 0. 05), 细胞克隆形成数和侵袭细胞数减 少 (P< 0. 05), 且呈剂量依赖性; 与 miR-NC 组比较, miR-4429 组细胞增殖抑制率和 E-cadherin 蛋白水平升 高 (P< 0. 05), 细胞克隆形成数和侵袭细胞数减少 (P< 0. 05), 划痕愈合率和 N-cadherin 蛋白水平降低 (P < 0. 05); 与 TFV + anti-miR-NC 组比较, TFV + anti-miR-4429 组细胞增殖抑制率和 E-cadherin 蛋白水平降低 (P<0. 05), 细胞克隆形成数和侵袭细胞数增多 (P<0. 05), 划痕愈合率和 N-cadherin 蛋白水平升高 (P< 0. 05)。 结论 蜘蛛香总黄酮可抑制胃癌 AGS 细胞增殖、 迁移及侵袭, 其抗肿瘤机制与上调 miR-4429 表达有关。

Abstract: Objective To explore the effect of total flavonoids from Valeriana jatamansi Jones (TFV) on gastric cancer cells AGS proliferation, migration and invasion and its possible mechanism. Methods In vitro cultured human gastric cancer cells AGS were randomly divided into Con group, TFV-L group, TFV-M group, TFV-H group, miR-NC group, miR-4429 group, TFV + anti-miR-NC group, and TFV + anti-miR-4429 group. The expression of miR-4429, cell proliferation, clone formation ability, cell migration and invasion were detected by qRT-PCR, CCK-8, plate colony formation assay, wound healing assay and transwell assay, respectively. The expression levels of E-cadherin and N-cadherin proteins were detected by Western blotting. Results Compared with the Con group, the expression levels of E-cadherin, proliferation inhibition rate and miR-4429 expression level in the TFV-L, TFV-M, and TFV-H groups were increased (P< 0. 05), while the expression level of N-cadherin and the wound healing rate were decreased (P< 0. 05), the number of colonies formed and the number of invasive cells were decreased (P < 0. 05), all in a dose-dependent manner. Compared with the miR-NC group, the cell proliferation inhibition rate and the expression level of E-cadherin in the miR-4429 group were increased (P< 0. 05), while the number of cell clones and the number of invasive cells were decreased (P< 0. 05), the wound healing rate and the expression level of N-cadherin were decreased (P < 0. 05). Compared with the TFV + anti-miR-NC group, the cell proliferation inhibition rate and the expression level of E-cadherin in the TFV + anti-miR-4429 group were decreased (P< 0. 05), while the number of colonies formed and the number of invasive cells were increased (P< 0. 05), the wound healing rate and the expression level of N-cadherin were increased (P< 0. 05). Conclusion Total flavonoids from Valeriana jatamansi Jones can inhibit gastric cancer AGS cells proliferation, migration and invasion, and its anti-tumor mechanism may be related to the up-regulation of miR-4429 expression.
