摘要: 目的 探究 ω-3 多不饱和脂肪酸 (ω-3 polyunsaturated fatty acids, ω-3 PUFAs) 对慢性难愈性创面大鼠创面愈合的影响方法 SD 大鼠随机分为正常组模型组、 ω-3 PUFAs 高剂量组和贝复新组, 每组 10 , 除正常组外其余各组大鼠构建慢性难愈性创面模型, ω-3 PUFAs 和贝复新分组处理后检测大鼠创面愈合比例; HE 染色检测创面组织形态结构; 免疫组化染色检测创面组织阳性 CD31 表达来评测其微血管密度 (microvascular density, MVD); ELISA 法检测血清促炎因子 TNF-α、 IL-6 水平; 免疫组化染色检测创面组织 Collagen Ⅱ 、 纤维结合蛋白 ( fibrin-binding protein, FN) 阳性表达; 蛋白质印迹检测创面组织促炎因子与 VEGF、 TGF-β1 蛋白表达结果 模型组大鼠 TNF-α IL-6 水平、 MVD、 Collagen Ⅱ FN阳性表达、 TNF-α IL-6、 VEGF、 TGF-β1 蛋白表达相比正常组均升高 (P< 0. 05); 相比模型组, ω-3 PUFAs 高剂量组与贝复新组 TNF-α IL-6 水平、 TNF-α IL-6 蛋白表达降低, 创面愈合比例、MVD、 Collagen Ⅱ FN 阳性表达、 VEGF TGF-β1 蛋白表达升高 (P< 0. 05), ω-3 PUFAs 各剂量组间各指标变化呈剂量依赖性 (P< 0. 05); 贝复新组各指标相比 ω-3 PUFAs 高剂量组无显著变化 ( P > 0. 05)。 结论 ω-3 PUFAs 可抑制创面炎症促进其血管生成, 并上调 VEGF、 TGF-β1 表达, 进而促使慢性难愈性创面愈合

Abstract: Objective To investigate the effect of ω-3 polyunsaturated fatty acids ( ω-3 PUFAs) on wound healing in rats with chronic refractory wounds. Methods SD rats were randomlyseparated into 6 groups: normal group, model group, ω-3 PUFAs low-, medium-, and high-dose groups, and Beifuxin group, with 10 rats in each group. Except for the normal group, the rats in the other groups were constructed with a chronic refractory wound model, and the wound healing ratio of the rats was measured after being treated with ω-3 PUFAs or Beifuxin. HE staining was used to determine the morphological structure of wound tissues. Immunohistochemical staining was used to detect the expression of Collagen Ⅱ and fibrin-binding protein (FN) and the expression of CD31 in the wound tissues to evaluate their microvascular density (MVD). ELISA detected the serum levels of pro-inflammatory factors TNF-α and IL-6. Western blotting was used to detect the expression levelsof pro-inflammatory factors and VEGF and TGF-β1 proteins in wound tissues. Results The levels ofTNF-α and IL-6, MVD, positive expressions of Collagen Ⅱ and FN, and the expression levels of TNF-α and IL-6, VEGF, TGF-β1 proteins in the model group were higher than those in the normalgroup (P < 0. 05). Compared with those in the model group, the levels of TNF-α and IL-6 and theprotein expression levels of TNF-α and IL-6 in the ω-3 PUFAs low-, medium-and high-dose groups and the Beifuxin group were decreased, the wound healing ratio, MVD, positive expressions ofCollagen Ⅱ and FN, the protein expression levels of VEGF and TGF-β1 were increased ( P < 0. 05). The changes in each index among ω-3 PUFAs groups were dose-dependent (P < 0. 05). No significant change was seen when compared those indexes between the Beifuxin group and the ω-3PUFAs high-dose group ( P > 0. 05). Conclusion  ω-3 PUFAs can inhibit wound inflammation,promote angiogenesis, and upregulate the expression of VEGF and TGF-β1, and promote the healing of chronic refractory wounds.
