CircITCH Regulates Proliferation and Invasion of Colorectal Cancer Cells via Targeting miR-106b #br#

摘要: 目的 研究环状 RNA circITCH 靶向微小 RNA ( miR) -106b 对结直肠癌细胞增殖及侵袭的调控作用方法 培养结直肠癌细胞株 SW620、 HT-29、 HCT116、 Caco-2、 SW480 及正常结肠上皮细胞株NCM460, 检测 circITCH miR-106b 的表达水平并筛选表达变化最显著的 HT-29 细胞进行分组: 阴性对照(NC) 质粒组, circITCH 质粒组, miR-NC , miR-106b , NC 质粒 + miR-NC , circITCH 质粒 + miR-NC, circITCH 质粒 + miR-106b 质粒转染 48 h 后检测细胞增殖的吸光度值 ( A450 nm )、 侵袭数目以及PTEN、, BTG3 (b-cell translocation gene 3) 蛋白的表达水平结果 相比于 NCM460 细胞, SW620、 HT-29、HCT116、 Caco-2、 SW480 细胞中 circITCH 的表达水平较低, 以上细胞中 miR-106b 的表达水平均高于NCM460 细胞中表示水平 (P< 0. 05)。 miR-106b 组中野生型 circITCH 报告基因的荧光素酶活性低于 miR-NC(P< 0. 05), 突变型 circITCH 报告基因的荧光素酶活性与 miR-NC 组比较无显著性差异 (P > 0. 05)。 circITCH 质粒组 HT-29 细胞中 circITCH、 PTEN、 BTG3 的表达水平高于 NC 质粒组, miR-106b 的表达水平及A450侵袭数目均低于 NC 质粒组 (P< 0. 05)。 circITCH 质粒 + miR-106b HT-29 细胞中 miR-106b 的表达水平及 A450 nm侵袭数目均高于 circITCH 质粒 + miR-NC , PTEN、 BTG3 的表达水平低于 circITCH 质粒 +miR-NC (P< 0. 05)。 结论 circITCH 抑制结直肠癌细胞增殖及侵袭, 靶向 miR-106b 是相关的分子机制

Abstract: Objective To study the effect of circITCH on the proliferation, invasion of colorectal cancer cells via targeting miR-106b. Methods Colorectal cancer cell lines SW620, HT-29,HCT116, Caco-2, SW480, and normal colon epithelial cell line NCM460 were cultured. The expression levels of circITCH and miR-106b were detected, and HT-29 cells were selected for further experiments and were divided into 7 groups: Negative control ( NC) group, circITCH group, miR-NC group, miR-106b group, NC plasmid + miR-NC group, circITCH + miR-NC group, andcircITCH + miR-106b group. The A450 value, number of invasion cells and the expression levels of PTEN and B cell translocation gene 3 (BTG3) were detected. Results The expression level of circITCH in the SW620, HT-29, HCT116, Caco-2 and SW480 cells was lower than that in theNCM460 cell, while the expression level of miR-106b was higher in the NCM460 cell (P < 0. 05).Dual-luciferase reporter gene assay showed that miR-106b could bind with circITCH reporter gene). The expression levels of circITCH, PTEN, BTG3 in the circITCH group were higher than those inthe NC group, the expression level of miR-106b, A450 value, and the number of invasion cells were lower than those in the NC group (P < 0. 05). The expression level of miR-106b, A450 value, andthe number of invasion cells in the circITCH + miR-106b group were higher than those in the circITCH + miR-NC group, the expression levels of PTEN, BTG3 were lower than those in the circITCH + miR-NC group (P< 0. 05). Conclusion CircITCH can inhibit the proliferation and invasion of colorectal cancer cells via targeting miR-106b.
