Effect of Compound Shougong Powder on Pain Sensitization in Rats with Bone Cancer Pain through TNF-α Signaling Pathway

摘要: 目的 探讨复方守宫散 (Compound Shougong powder, CSP) 通过肿瘤坏死因子 α (TNF-α) 信号 通路对骨癌痛大鼠痛觉敏化机制的影响。 方法 通过注射 Lewis 肺癌细胞建立肺癌骨癌痛大鼠模型; 将大 鼠随机分为模型组、 阳性对照 (扶他林, 0. 1 g / kg) 组、 CSP 低 (CSP-L, 20 mg / kg)、 中 (CSP-M, 40 mg / kg)、 高 (CSP-H, 80 mg / kg) 剂量组, 并设置假手术组大鼠, 每组各 10 只。 干预结束后, 检测大鼠机械 性缩足反射阈值 (paw withdrawal mechanical threshold, PWMT); 全自动热痛测试仪检测大鼠热痛觉缩足潜 伏期 (paw withdrawal thermal lathency, PWTL); ELISA 法检测血清中 TNF-α、 白介素 ( IL) -1β、 IL-6 水平 变化; Western 印迹检测各组大鼠脊髓组织单核细胞趋化蛋白-1 (MCP-1) 以及 TNF-α 表达水平。 结果 与 假手术组相比, 模型组大鼠 IL-1β、 IL-6 水平、 TNF-α、 MCP-1 表达均显著升高, PWMT 显著降低, PWTL 显著缩短 ( P < 0. 05); 与模型组相比, CSP-L 组、 CSP-M 组、 CSP-H 组大鼠 IL-1β、 IL-6 水平、 TNF-α、 MCP-1 表达均显著降低, PWMT 显著升高, PWTL 显著延长 (P< 0. 05); CSP-H 组各项指标与阳性对照组 差异无统计学意义 (P> 0. 05)。 结论 CSP 可以改善肺癌骨癌痛大鼠疼痛反应, 发挥止痛作用, 可能与抑 制 TNF-α 信号通路有关。

Abstract: Objective To investigate the effect of Compound Shougong powder (CSP) on the pain sensitization in rats with bone cancer pain through tumor necrosis factor-α (TNF-α) signaling pathway. Methods A rat model of lung cancer bone metastases pain was established by injecting Lewis lung cancer cells. Rats were randomly divided into 6 groups: model group, positive control group ( Voltaren, 0. 1 g / kg), CSP low-dose group ( CSP-L, 20 mg / kg), CSP medium-dose group (CSP-M, 40 mg / kg), CSP high-dose group (CSP-H, 80 mg / kg), and the sham operation group, 10 rats in each group. After the intervention, the paw withdrawal mechanical threshold (PWMT) of the rats was detected, and the paw withdrawal thermal latency (PWTL) in rats was detected by the automatic thermal pain tester. The levels of TNF-α, IL-1β and IL-6 in serum were detected by ELISA. Western blotting was performed to detect the expression levels of monocyte chemoattractant protein-1 (MCP-1) and TNF-α in spinal cord tissues of rats in each group. Results The levels of IL-1β, IL-6, TNF-α and MCP-1 in the model group were significantly increased when compared with those in the sham operation group, while the values of PWMT and PWTL were significantly decreased (P<0. 05). The levels of IL-1β, IL-6, TNF-α and MCP-1 in the CSP-L group, CSPM group and CSP-H group were significantly decreased when compared with those in the model group, while the values of PWMT and PWTL were significantly increased (P<0. 05). No obvious differences in the above indexes were found between the CSP-H group and the positive control group (P > 0. 05). Conclusion CSP may alleviate the pain response in rats with lung cancer bone metastases pain and exert analgesic effect by inhibiting the expression of TNF-α and other inflammatory factors.
