摘要: 周围神经病是周围神经系统结构或功能受损的一类疾病,其病因十分复杂,常由创伤、高血糖、化疗药物副作用以及炎症、免疫相关、代谢、内分泌相关、肿瘤等因素诱发,给患者带来极大痛苦。氧化应激作为产生周围神经病的关键原因之一,主要表现为体内氧化过程与抗氧化防御失衡,倾向于氧化,导致中性粒细胞炎性浸润,蛋白酶分泌增加,产生大量氧化中间产物。目前临床上广泛应用抗氧化剂来抑制氧化应激反应,改善周围神经病,但疗效有限。核因子红系2相关因子2(nuclear factor-erythroid 2-related factor 2,Nrf2)作为细胞内氧化应激的关键转录因子,通过众多信号通路,介导抗氧化作用,如Nrf2/HO-1、Nrf2/NQO1、PERK/Nrf2、Nrf2/ERK、Nrf2/NF-κB等。该文通过综述靶向细胞Nrf2因子相关的氧化应激通路,以期为周围神经病相关研究提供证据。

Abstract: Peripheral neuropathies are a collection of disorders characterized by injury and generalized dysfunction of nerves.Pathogenic factors are complex,including trauma,hyperglycemia,side effects of chemotherapeutic drugs and inflammatory,immune-related,metabolic,endocrine-related,and neoplastic factors.In addition,it can cause great problems for patients.Oxidative stress,as one of the key causes of peripheral neuropathy,is characterized mainly by an imbalance between oxidative processes and antioxidant defenses in the body,which tends to oxidize,leading to inflammatory infiltration of neutrophils,increased protease secretion,and the production of many oxidative intermediates.At present,antioxidants are widely used in clinical practices to inhibit oxidative stress to improve peripheral neuropathy,but their efficacy is limited.Nuclear factor-erythroid 2-related factor 2(Nrf2),a key transcription factor of intracellular oxidative stress,mediates antioxidant effects through numerous signaling pathways,such as the Nrf2/HO-1,Nrf2/NQO1,PERK/Nrf2,Nrf2/ERK,and Nrf2/NF-κB pathways.In the present study,the oxidative stress pathways associated with the targeting of cellular Nrf2 factors were reviewed,hopefully providing evidence for peripheral neuropathy-related research.
