摘要: 目的 探讨多囊卵巢综合征患者 (polycystic ovary syndrome, PCOS) 卵巢颗粒细胞 miR-144-3p 及其靶基因 PTEN 的表达及作用 方法选取 2020 10 月至 2022 3 月在淮安市妇幼保健院生殖医学科行IVF / ICSI 治疗的 20 PCOS 患者作为研究对象即为 PCOS , 选取行 IVF / ICSI 治疗的输卵管因素或男方因素患者 20 例作为对照组收集两组患者颗粒细胞, 并分析颗粒细胞 miR-144-3p 的表达情况利用 TargetScan 数据库预测 miR-144-3p 的靶基因, 并采用双荧光素酶活性检测验证靶基因复苏人卵巢颗粒细胞(ovarian granulosa cells, KGN), 转染并建立 miR-144-3p 模拟物组模拟物阴性对照组、 miR-144-3p 抑制物组抑制物阴性对照组、 si-PTEN siRNA-NC 采用流式细胞仪检测各组的细胞凋亡情况, RT-PCR 及蛋白质印迹技术检测颗粒细胞中 miR-144-3p、 PTEN mRNA 及蛋白的表达情况结果 RT-PCR 结果显示 PCOS miR-144-3p 表达水平显著低于对照组 (P< 0. 05), 流式细胞仪检测结果显示 miR-144-3p mimic 组的颗粒细胞凋亡百分比显著低于 mimic-NC (P< 0. 05), miR-144-3p inhibitor 组颗粒细胞凋亡水平显著高于inhibitor-NC (P< 0. 05)。 生物信息学预测 miR-144-3p 的靶基因为 PTEN, 荧光素酶结合实验证实 miR- 144-3p 能特异结合 PTEN-3′UTR 并下调 PTEN 基因表达。 RT-PCR 测定结果显示 PCOS PTEN mRNA 表达显著高于对照组 (P< 0. 05), 且与 miR-144-3p 表达水平呈负相关 (r = - 0. 91, P< 0. 001)。 qRT-PCR 及蛋白质印迹检测结果显示与 mimic NC 组相比, miR-144-3p mimic 组中 PTEN mRNA 及蛋白表达量显著低于mimic NC (P< 0. 05)。 miR-144-3p inhibitor PTEN mRNA 及蛋白表达量显著高于 inhibitor NC (P< 0. 05)。 流式细胞仪检测结果显示 si-PTEN 组颗粒细胞凋亡水平显著低于 siRNA-NC (P< 0. 05), miR- 144-3p inhibitor + si-PTEN 组的颗粒细胞凋亡比例显著低于 miR-144-3p inhibitor (P< 0. 05)。 结论 PCOS患者卵巢颗粒细胞 miR-144-3p 表达水平显著降低, PTEN 基因表达水平显著增高, 进而促进 PCOS 患者卵巢颗粒细胞凋亡

Abstract: Objective To investigate the expressions of miR-144-3p and its target gene PTEN in ovarian granule cells and their functions in patients with polycystic ovary syndrome. Methods Twenty patients with PCOS and 20 controls were recruited. Granulosa cells were collected in both groups and the expression level of miR-144-3p in cells was analyzed. The target genes of miR-144-3p were predicted by TargetScan database, and the binding relationship was verified by dual luciferase gene reporter assay. The human ovarian granulosa cells ( KGN) were resuscitated and transfected with miR-144-3p mimic / mimic NC, or miR-144-3p inhibitor / inhibitor NC, or si-PTEN / siRNANC, and were then accordingly divided into 6 groups: miR-144-3p mimic group, mimic NC group, miR-144-3p inhibitor group, inhibitor NC group, si-PTEN and siRNA-NC group. Cell apoptosis was detected by TUNEL assay. RT-PCR and Western blotting method were used to detect the relative expression levels of miR-144-3p and PTEN. Results RT-PCR results showed that the expression level of miR-144-3p in the PCOS group was significantly lower than that in the control group(P< 0. 05). Flow cytometry results showed that the apoptosis rate of granulosa cells in the miR-144- 3p mimic group was significantly lower than that in the mimic-NC group (P< 0. 05), while the apoptosis rate of granulosa cells in the miR-144-3p inhibitor group was significantly higher than that inthe inhibitor-NC group (P< 0. 05). PTEN was predicted as the target gene of miR-144-3p by bioinformatics analysis, and the luciferase gene reporter experiments had confirmed that miR-144-3pcould specifically bind to the PTEN-3 ′ UTR and down-regulate the expression level of PTEN gene. RT-PCR assay results showed that the expression level of PTEN mRNA in the PCOS group was significantly higher than that in the control group (P < 0. 05), and was negatively correlated with the expression level of miR-144-3p (r = - 0. 91, P< 0. 001). The results of qRT-PCR and Westernblotting showed that the mRNA and protein expression levels of PTEN in the miR-144-3p mimicgroup were significantly lower than those in the NC mimic group (P< 0. 05), while the mRNA andprotein expression levels of PTEN in the miR-144-3p inhibitor group were significantly higher thanthose in the inhibitor NC group (P< 0. 05). Flow cytometry showed that the apoptosis rate of granulosa cells in the si-PTEN group was significantly lower than that in the siRNA-NC group ( P <0. 05), while the apoptosis rate of granulosa cells in the miR-144-3p inhibitor + si-PTEN group wassignificantly lower than that in the miR-144-3p inhibitor group (P < 0. 05). Conclusion The expression level of miR-144-3p in ovarian granule cells in PCOS patients is significantly reduced,while the expression level of PTEN gene is significantly increased, which may have promoted theapoptosis of ovarian granule cells in PCOS patients.
