热毒宁注射液对肺炎克雷伯菌所致幼龄小鼠肺功能损伤和免疫紊乱的调节作用

Effects of Reduning Injection on Lung Function Injury and Immune Disorder Induced by Klebsiella Pneumoniae in Young Mice

摘要: 目的 探讨热毒宁注射液对肺炎克雷伯菌诱导的幼龄小鼠肺功能损伤和免疫紊乱调节的影响。 方法 将小鼠随机分为 5 组: 对照组、 模型组、 热毒宁注射液低中高剂量组。 HE 染色检测肺组织病理情 况。 肺组织病理学评分和 W/ D 值评估肺损伤。 RT-PCR 检测 Ki67、 Survivin mRNA 表达。 Westernt 印迹检测 Caspase-3、 Caspase-9、 TGF-β 和 α-SMA 蛋白表达。 Masson 染色检测肺组织纤维化。 ELISA 检测肺组织和外 周血中 iNOS、 IL-10 含量。 结果 与模型组比较, 热毒宁注射液中高剂量组的肺损伤和肺纤维化明显改善, 肺组织病理学评分和 W/ D 值降低 (P< 0. 05), Ki67 和 Survivin mRNA 表达升高 (P< 0. 05), Caspase-3、 Caspase-9、 TGF-β 和 α-SMA 蛋白表达降低 (P< 0. 05), 组织和外周血中的 iNOS 含量降低 (P< 0. 05), IL-10 含量升高 (P< 0. 05)。 结论 热毒宁注射液可改善肺炎克雷伯菌所致幼龄小鼠的肺功能损伤和免疫紊 乱。

Abstract: Objective To investigate the effects of Reduning injection on lung function injury and immune disorder induced by Klebsiella pneumoniaein young mice. Methods The mice were randomly divided into 5 groups: control group, model group and low-, medium-and high-dose Reduning injection groups. HE staining was used to detect the pathology of lung tissues. Lung histopathological score and W/ D ratio were used to assess lung injury. The expression of Ki67 and Survivin mRNA was detected by RT-PCR. The expression of Caspase-3, Caspase-9, TGF-β and α-SMA proteins were detected by Western blot. Masson staining was used to determine pulmonary fibrosis. The contents of iNOS and IL-10 in lung tissue and peripheral blood were determined by ELISA. Results In Reduning injection groups relative to the KPN group, the lung injury and pulmonary fibrosis were significantly improved; the lung tissue score and W/ D ratio were markedly decreased (P < 0. 05) . The expression levels of Ki67 and Survivin mRNA were significantly increased (P< 0. 05). The expression levels of Caspase-3, Caspase-9, TGF-β and α-SMA protein were profoundly decreased (P< 0. 05). The iNOS content in tissues and peripheral blood was significantly decreased ( P < 0. 05). The IL-10 content was obviously increased (P < 0. 05). Conclusion Reduning injection can alleviate the lung function injury and immune disorder caused byKlebsiella pneumoniaein young mice.
