抑制下丘脑 TLR4 表达对肥胖相关代谢状态的影响

Effect of Inhibition of TLR4 Expression in Hypothalamus on Obesity-Related Metabolic States

摘要: 目的 研究抑制下丘脑 TLR4 表达对肥胖相关代谢状态的影响。 方法 40 只 4 周龄雄性 C57BL/ 6 小鼠 (SPF 级), 适应性喂养 1 周后, 随机分成 2 组, 分别饲喂正常饮食 (NCD)、 高脂饮食 (HFD) 4 周, 喂养期间, 每周测量体重 1 次。 9 周龄时, shRNA-TLR4 慢病毒颗粒及对照病毒颗粒分别注射正常饮食组和 高脂饮食组小鼠。 13 周龄时处死小鼠, 并采用实时荧光定量 qPCR 方法, 检测 TLR4 在下丘脑中的表达水 平; 血糖仪检测小鼠空腹血糖; ELISA 法检测血清 TNF-α、 IL-6; 应用自动生化分析仪检测血清低密度脂蛋 白-胆固醇、 高密度脂蛋白-胆固醇、 甘油三脂及总胆固醇水平; 脂肪组织 HE 染色。 结果 TLR4-shRNA 慢 病毒干预后, 下丘脑组织中 TLR4 的表达显著减少, 差异有统计学意义 (P< 0. 05); 高脂喂养的 C57BL/ 6 小鼠的体重增长量、 能量摄入量均明显降低, 差异有统计学意义 (P< 0. 05); 高脂喂养的 C57BL/ 6 小鼠的 炎性因子、 甘油三酯、 空腹血糖明显下降, 差异有统计学意义 (P< 0. 05)。 脂肪细胞肥大现象缓解。 结论 通过有效干预下丘脑组织中 TLR4 的表达水平, 可显著改善肥胖相关的炎性反应、 糖、 血脂等代谢紊乱。

Abstract: Objective To investigate the effect of inhibiting the expression of TLR4 in the hypothalamus on obesity-related metabolic state. Methods After 1-week adaptive feeding, 40 four week-old male C57BL / 6 mice (SPF grade) were randomly divided into two groups, in which the mice were fed with normal diet (in NCD group) or high-fat diet (in HFD group) for 4 weeks respectively. The body weight of mice were measured once a week. The mice in each of the above groups were then randomly selected and divided into two group for the injection of TLR4 shRNA lentiviral particles or control viral particles respectively. Mice were sacrificed when reached 13-week old for further experiments. The expression of TLR4 in the hypothalamus was detected by fluorescence quantitative RT-PCR. The fasting blood glucose of the mice was detected by a blood glucose meter. The levels of serum TNF-α and IL-6 were detected by ELISA. The levels of serum low density lipoprotein-cholesterol, high density lipoprotein-cholesterol, triglyceride and total cholesterol were detected by automatic biochemical analyzer. HE staining were performed in adipose tissues. Results After TLR4-shRNA lentivirus intervention, the expression level of TLR4 in the hypothalamus was significantly decreased (P< 0. 05), and the weight gain and energy intake of high-fat fed C57BL / 6 mice were significantly decreased (P < 0. 05), the levels of inflammatory factors, triglycerides, and fasting blood glucose were significantly decreased in high-fat fed C57BL / 6 mice (P < 0. 05). The hypertrophy of adipocyte was also alleviated after TLR4-shRNA lentivirus intervention. Conclusion Down-regulation of TLR4 expression in the hypothalamus can effectively improve obesity-related inflammatory response, and metabolic disorders on glucose and blood lipids.
