Expression of CDC40 in Colorectal Cancer and Its Correlation with Clinicopathological Features

Abstract: Objective To investigate the expression of CDC40 in colorectal cancer tissues and its correlation with the clinicopathological features,so as to provide a basis for the early diagnosis,targeted therapy and prognostic assessment of colorectal cancer. Methods Immunohistochemical staining was used to evaluate the expression level of CDC40 in the tissues of 70 colorectal cancer patients,and the relationship between the CDC40 expression and the clinicopathological features was analyzed.The R software package was used for survival curve and multivariate prognostic analysis.Cell culture,transfection of CDC40 plasmid,proliferation experiments,Western blotting and nude mouse tumorigenesis experiments were conducted by using the colorectal cancer cell lines SW620 and Caco-2 to verify the effect of CDC40 on cell proliferation.The experimental data were statistically analyzed using IBM SPSS Statistics Version 22 software. Results In 70 patients,the CDC40 high-expression group accounted for 51.4 %.The expression level of CDC40 was related to the tumor differentiation degree,T stage,lymph node metastasis and the levels of Ki67,but not to gender,age and distant metastasis.Multivariate and univariate survival analyses showed that the high expression of CDC40 was associated with poorer survival prognosis of patients.In vitro experiments showed that overexpression of CDC40 promoted the proliferation of colorectal cancer cells.The nude mouse tumorigenesis experiment demonstrated that the overexpression of CDC40 promoted the tumor growth. Conclusion CDC40 plays an important role in the occurrence and development of colorectal cancer and can be used as a potential molecular marker and therapeutic target.
