Effects of Reduning Injection on Lung Function Injury and Immune Disorder Induced by Klebsiella Pneumoniae in Young Mice

Abstract: Objective To investigate the effects of Reduning injection on lung function injury and immune disorder induced by Klebsiella pneumoniaein young mice. Methods The mice were randomly divided into 5 groups: control group, model group and low-, medium-and high-dose Reduning injection groups. HE staining was used to detect the pathology of lung tissues. Lung histopathological score and W/ D ratio were used to assess lung injury. The expression of Ki67 and Survivin mRNA was detected by RT-PCR. The expression of Caspase-3, Caspase-9, TGF-β and α-SMA proteins were detected by Western blot. Masson staining was used to determine pulmonary fibrosis. The contents of iNOS and IL-10 in lung tissue and peripheral blood were determined by ELISA. Results In Reduning injection groups relative to the KPN group, the lung injury and pulmonary fibrosis were significantly improved; the lung tissue score and W/ D ratio were markedly decreased (P < 0. 05) . The expression levels of Ki67 and Survivin mRNA were significantly increased (P< 0. 05). The expression levels of Caspase-3, Caspase-9, TGF-β and α-SMA protein were profoundly decreased (P< 0. 05). The iNOS content in tissues and peripheral blood was significantly decreased ( P < 0. 05). The IL-10 content was obviously increased (P < 0. 05). Conclusion Reduning injection can alleviate the lung function injury and immune disorder caused byKlebsiella pneumoniaein young mice.
