摘要:目的 采用网络药理学的方法探究灯银脑通胶囊(DYNT)治疗脑卒中的有效成分群及关键靶点预测。方法 基于中药系统药理学数据库与分析平台(TCMSP)、中医药研究综合数据库(TCMID)和 TCM-Mesh,收集 DYNT的化学 成分,并通过 DrugBank、毒性与基因比较数据库(CTD)、STITCH、BindingDB和SwissTarget等数据库预测 DYNT各化 学成分的靶点数据。在 DisGeNET、PharmGKB和 CTD 数据库中检索脑卒中相关靶点,取疾病靶点和 DYNT 成分相关 靶点交集作为 DYNT作用脑卒中的潜在靶点。通过 STRING 数据库建立蛋白质相互作用(PPI)网络,使用 MCODE cluster识别 DYNT的核心靶点。运用 CTD数据库对 DYNT治疗脑卒中相关靶点及调控脑卒中相关通路进行 GO 富集 分析和 KEGG信号通路富集分析。利用 MCC 和 Stress两种算法识别网络中排名前十的关键靶点,取两者交集作为 DYNT治疗脑卒中的关键靶点。结果 从 DYNT中共收集到133个化学成分,涉及靶点588个,化学成分中木犀草素、 山奈酚和芹菜素等59个成分参与了对脑卒中的调控。DYNT 治疗脑卒中的靶点共有 153个,涉及通路 12个,基于 MCC和Stress两种算法发现 DYNT治疗脑卒中的8个关键靶点。结论 DYNT 中的各化学成分可能通过 PI3K-Akt、 HIF-1和 Toll样受体等12条信号通路发挥对脑卒中的调控作用,AKT1、MAPK3、TP53、IL6、VEGFA、TNF、INS和 ALB可能为 DYNT治疗脑卒中的8个关键靶点。
